| I. Background | |

血小板衍生生长因子（PDGF）是一类重要的生长因子，主要由血小板、内皮细胞和 巨噬细胞分泌，在细胞增殖、迁移、分化和血管生成等生物学过程中发挥关键作用。它不 仅在正常生理过程中（如伤口愈合和组织修复）起重要作用，还在多种病理过程中（如癌 症、纤维化和动脉粥样硬化）扮演重要角色。PDGF 家族包括 PDGF-A、PDGF-B、PDGF- C 和 PDGF-D 四种亚型。PDGF 通过与其受体（PDGFR）结合，激活下游信号通路，调控 细胞行为。 血小板衍生生长因子受 0 体α（PDGFRA），又称为 CD140A 或 PDGFR2，是一种细胞 表面酪氨酸激酶受体，属于血小板衍生生长因子家族。PDGFRA 与配体 PDGF 结合后会二 聚化以及磷酸化，激活包括 JAK-STAT3、PI3K-AKT-mTOR 和 RAS-MAPK 等下游信号通路， 在调节关键细胞功能如增殖和凋亡方面有重要作用。PDGFRA 蛋白被激活异常时，则会导 致肿瘤的发生并促进肿瘤血管生成。 |

PDGF/PDGFRA Effector Reporter Cell 报告基因药靶模型很好的模拟了体内 PDGFRA 的信号转导过程，原理见下图所示。
